Instructions for using a bacterial collection instrument for sterile testing of liquid samples

1. Take out the incubator and check if the packaging is intact. Open the plastic packaging bag in the sterile room.

2. Insert the incubators one by one onto the stainless steel seats.

3. Install the elastic hose of the bacterial culture incubator into the pump head of the intelligent bacterial collection device, ensuring accurate positioning and smooth movement of the hose.

4. Open the bottle cap insertion hole of the sample to be tested and disinfect it (if testing ampoule samples, disinfect the neck of the ampoule first, then open and position the ampoule).

5. Remove the sheath of the dual core injection tube, insert it into the sample bottle, turn on the bacterial collector, and perform filtration and bacterial collection (avoid air intake into the dual core injection tube, as the filter membrane may be soaked by the drug solution and affect air intake).

6. After completing the bacterial collection, if the sample contains antibacterial substances, it should be washed with appropriate rinsing solution according to the pharmacopoeia specifications, and the cleaning method should be the same as the bacterial collection process.

7. Disinfect the insertion hole of the culture medium bottle.

8. Remove the rubber plug from the top air filter opening and place it on the bottom of the incubator. Use a hose clamp to sequentially open and close the hose, turn on the bacterial collector, and pump the culture medium into the incubator.

9. Use a clip to clamp the hose connected to the incubator, leaving a 5-6cm hose, cut off the remaining part, and insert the open end into the air filter opening.

10. Cultivate according to regulations separately.

11. Observe the cultivation situation. If sampling and separation cultivation are required, disinfect the hose and insert sterile injection into the hose to extract the required amount for further inspection.

Method of using microbial limit testing (repeated use of culture equipment)

Membrane filtration method for purified water, injection water, and supernatant

1. Take N filter membranes (diameter 50mm) and soak them in purified water for about 5 minutes

2. Use tweezers to remove the filter membrane and place it flat on the stainless steel mesh. Place the stainless steel mesh into the stainless steel base, insert the gasket and O-ring, and tightly assemble the screw cover and cup cover to form a complete set of filter incubators.

3. Sterilization: Wrap the assembled incubator in kraft paper and place it in a high-pressure moist heat sterilization pot for sterilization (according to the regulations of the 2015 edition of the Chinese Pharmacopoeia).

4. Take the sterilized incubator to the microbiological limit testing room for bacterial collection and filtration.

5. Open the configured solid culture medium and place the collected filter membrane flat on the culture medium (no bubbles should be generated on the filter membrane)

6. Cultivate according to the regulations of the Chinese Pharmacopoeia.
